Cd31 Cd34 Markers

CD31 and CD34 markers are cell-surface proteins used to identify and characterize distinct cell populations in biological samples. CD31, also called PECAM-1, supports cell adhesion and signaling at endothelial cell junctions, whereas CD34 is a transmembrane sialomucin associated with hematopoietic progenitor and endothelial cell populations. Researchers detect these markers with antibody-based methods such as flow cytometry, immunohistochemistry, and immunofluorescence, using their expression patterns to distinguish cells and assess tissue organization. In biology, CD31 and CD34 markers help investigate blood vessel formation, stem and progenitor cells, vascular development, tissue repair, and disease-related changes in the vascular system.

The differentiation of white and beige adipocytes from adipose tissue vascular progenitors bears potential for metabolic improvement in obesity. We describe protocols for a CD34+CD31+ endothelial cell isolation from human fat and for a subsequent in vitro expansion and differentiation into white and beige adipocytes. Several downstream applications are discussed.

Liver injuries are accompanied by progenitor cell expansion that represents a heterogeneous cell population. Novel classification of this cellular compartment allows for the distinguishing of multiple subsets. The method described here illustrates the flow cytometry analysis and high purity isolation of various subsets that can be used for further assays.

This study has established a stable and efficient method for the isolation, culture, and functional determination of vascular wall-resident CD34+ stem cells (CD34+ VW-SCs). This easy-to-follow and time-effective isolation method can be utilized by other investigators to study the potential mechanisms involved in cardiovascular diseases.
