Chronic Infection Model

A chronic infection model is an experimental system that reproduces the long-term persistence of a pathogen, its components, or infection-associated host responses, helping researchers study disease beyond the acute phase. These models maintain sustained host–pathogen interactions in cells, tissues, organoids, or animals, where incomplete pathogen clearance, immune evasion, altered inflammation, and tissue remodeling can shape ongoing infection. In biology, chronic infection models support investigation of pathogen persistence, immune regulation, disease progression, and tissue damage. They also provide platforms for evaluating antimicrobial therapies, vaccines, and strategies designed to restore immune control or eliminate infection reservoirs.

Source: Lo, B. C., et al. Chronic Salmonella Infection Induced Intestinal Fibrosis. J. Vis. Exp. (2019)The video demonstrates the quantification of bacterial load in a mouse model of chronic infection with streptomycin-resistant Salmonella typhimurium. Following euthanasia and sterile dissection, cecal and splenic tissues are harvested, homogenized with steel beads to release bacteria, and serially diluted. Dilutions are plated on selective agar, and visible colonies are counted to estimate...

Source: de Vries, C. R., et.al. A Delayed Inoculation Model of Chronic Pseudomonas aeruginosa Wound Infection. J. Vis. Exp. (2020)This video demonstrates the delayed inoculation of luminescent Pseudomonas aeruginosa into a mouse bearing two full-thickness dorsal skin wounds covered by a transparent film dressing. This facilitates biofilm formation and the establishment of chronic infection. Regular bioluminescence monitoring enables quantification of bacterial burden and tracking of infection...
