Colony Assays

Colony assays are biological methods that measure the ability of individual cells to survive, proliferate, and form visible colonies under defined culture conditions. Cells are seeded at low density in a solid or semisolid medium, where each colony develops from a single cell or small cell cluster; researchers then count colonies or assess their size and morphology. These assays quantify clonogenic potential and can reveal how growth factors, genetic changes, drugs, or radiation affect cell survival and proliferation. In biology, colony assays support studies of stem and progenitor cells, cancer biology, microbial growth, and treatment response, providing a practical measure of cellular reproductive capacity.

In this video, we perform a clonogenic assay of keratinocytes harvested from adult mice to determine the ability of individual cells to proliferate and form colonies. This keratinocyte harvesting technique is useful for studying other downstream aspects of cutaneous chemical carcinogenesis and tumor promotion.

A Quantitative Assay for Insulin-expressing Colony-forming Progenitors

A three-dimensional clonogenic assay that allows pancreatic-like progenitors to differentiate into insulin-expressing colonies is described. This method takes advantage of semi-solid media containing methylcellulose, Matrigel and growth factors, in which single progenitors proliferate and differentiate in vitro, permitting quantification of the number of functional progenitors in a population.

The colony forming cell (CFC) assay is an in vitro assay in which hematopoietic progenitors form colonies in a semi-solid medium. A combination of colony morphology, cell morphology, and flow cytometry are used to assess the ability of the progenitors to proliferate and differentiate along the different hematopoietic lineages.
