Colony Picking

Colony picking is a microbiology technique used to select and transfer an individual microbial colony from a solid culture medium for further growth and analysis. After cells multiply on an agar plate, each visible colony typically represents a population derived from a single founding cell or cell cluster; a sterile tool or automated picker transfers the colony into fresh liquid medium or onto another plate. This process supports bacterial isolation, strain maintenance, recombinant DNA workflows, and screening for desired phenotypes or genetic constructs. Careful selection and aseptic handling help produce pure cultures, improve experimental reproducibility, and enable downstream identification, characterization, and biotechnology research.

In this video, we perform a clonogenic assay of keratinocytes harvested from adult mice to determine the ability of individual cells to proliferate and form colonies. This keratinocyte harvesting technique is useful for studying other downstream aspects of cutaneous chemical carcinogenesis and tumor promotion.

ScanLag: High-throughput Quantification of Colony Growth and Lag Time

ScanLag is a high-throughput method for measuring the delay in growth, namely lag time, as well as the growth rate of colonies for thousands of cells in parallel. Screening using ScanLag enables the discrimination between long lag-time and slow growth at the level of a single variant.
