Compound Uptake Efficacy

Compound uptake efficacy is the extent to which a biological system absorbs and retains a chemical compound, a key factor in determining its cellular activity and biological availability. Uptake can occur through passive diffusion across lipid membranes or through transporter-mediated processes and endocytosis, with efficiency influenced by compound properties, membrane permeability, transporter expression, and cellular conditions. Measuring uptake efficacy helps researchers compare compounds, characterize cell responses, and assess whether a candidate reaches its intended intracellular target. In biology and drug development, these evaluations support studies of pharmacological action, toxicity, delivery systems, and strategies for improving therapeutic performance.

2018

The protocol describes intubating adult zebrafish with a biologic; then dissecting and preparing the intestine for cytometry, confocal microscopy and qPCR. This method allows administration of bioactive compounds to monitor intestinal uptake and the local immune stimulus evoked. It is relevant for testing the intestinal dynamics of oral prophylactics.

Radiolabeled Amino Acid Uptake Assay to Quantify Cellular Uptake of Amino Acids

2025

This video demonstrates an in vitro technique for estimating amino acid uptake by bone marrow-derived stromal cells. Na+-dependent amino acid transporters mediate the uptake of radiolabeled amino acids inside the cells, and the cellular uptake is estimated by measuring the radioactivity.

Measuring Dopamine Uptake in Mouse Brain Synaptosomes Using Radiolabeled Dopamine

2025

This video demonstrates the assessment of dopamine uptake in synaptosomes harvested from mouse brains. It outlines steps for preparing synaptosomes for dopamine uptake via dopamine transporters, treating them with varying concentrations of radiolabeled dopamine, isolating the synaptosomes, and measuring radioactivity to quantify dopamine uptake.

Source: Kay Stewart, RVT, RLATG, CMAR; Valerie A. Schroeder, RVT, RLATG. University of Notre Dame, IN There are many commonly used routes for compound administration in laboratory mice and rats. However, certain protocols may require the use of less commonly used routes, including intradermal, intranasal, and intracranial injections. Specialized training is essential for these procedures to be performed successfully. Justification for these routes may need to be provided to gain Institutional...
