Connexin Proteins

Connexin proteins are membrane proteins that enable direct communication between neighboring cells, making them essential for coordinating activity in many tissues. Six connexin subunits assemble into a hemichannel, or connexon, and two connexons on adjacent cells dock to form a gap junction channel that permits ions and small signaling molecules to pass between cytoplasms. Connexins can also function as undocked hemichannels under specific conditions, influencing cell signaling and membrane behavior. Their selective permeability and regulated opening support electrical coupling, tissue development, metabolic coordination, and responses to injury, while connexin mutations or altered expression are associated with several human diseases.

Intercellular junctions are requisites for mammary gland stage-specific functions and development. This manuscript provides a detailed protocol for the study of protein-protein interactions (PPIs) and co-localization using murine mammary glands. These techniques allow for the investigation of the dynamics of the physical association between intercellular junctions at different developmental stages.

This video demonstrates a technique that uses biotinylated cell-penetrating peptides to detect intracellular protein-protein interactions. The cell-penetrating peptides facilitate the conjugated peptide to enter a cell and specifically bind to its interacting proteins. The biotin, after binding to avidin, allows purification of the interacting proteins’ complex from a cell lysate.

Rapid Assessment of Membrane Protein Quality by Fluorescent Size Exclusion Chromatography

The present protocol describes a procedure to perform fluorescent size exclusion chromatography (FSEC) on membrane proteins to assess their quality for downstream functional and structural analysis. Representative FSEC results collected for several G-protein coupled receptors (GPCRs) under detergent-solubilized and detergent-free conditions are presented.
