Cytokine Secretion Detection

Cytokine secretion detection is the measurement of signaling proteins released by cells, providing evidence of immune activation, communication, and inflammatory responses. In common immunoassays, cells are stimulated under controlled conditions, and secreted cytokines bind capture antibodies; a labeled detection antibody then produces a measurable signal proportional to cytokine abundance. Methods such as ELISA, ELISpot, and multiplex bead assays can quantify secretion in culture fluids or identify cytokine-producing cells. These measurements help characterize immune-cell function, compare cellular responses, evaluate inflammation, and assess effects of vaccines, drugs, or disease-related conditions in biological research.

Described here is a method for producing exogenous cytokine in patient-derived xenograft (PDX) mice via weekly intraperitoneal injection of a cytokine-transduced stromal cell line. This method broadens the utility of PDX and provides the option for transient or sustained exogenous cytokine delivery in a multitude of PDX models.

The protocol describes an advanced microfluidic platform to quantitatively measure cytokine secretion dynamics of individual human peripheral blood mononuclear cells.
