Estradiol Valerate

Estradiol valerate is a synthetic ester prodrug of estradiol, the principal biologically active estrogen, used to study and regulate estrogen-dependent biology. After administration, esterases hydrolyze estradiol valerate to release estradiol, which enters target cells, binds estrogen receptors, and alters gene transcription through receptor-mediated signaling. This activity influences reproductive tissues, bone maintenance, cardiovascular physiology, and other estrogen-responsive systems. In biology and medicine, estradiol valerate helps researchers investigate endocrine regulation and is used in hormone therapy and some contraceptive formulations, while its esterified structure can modify absorption and duration compared with unmodified estradiol.

Here, we describe how to identify the stage of the murine reproductive (proestrus, estrus, metestrus, or diestrus) by simple, non-invasive collection and cytological assessment of vaginal smear samples. We further describe how vaginal cytology reflects circulating hormonal levels underlying transition through the murine reproductive cycle.
