In Vitro Intestine

In vitro intestine refers to laboratory-grown models that reproduce key structural and functional features of the intestinal tract outside an organism, enabling controlled study of biology and disease. These systems use cultured intestinal cells, stem-cell-derived organoids, or microfluidic gut-on-chip platforms to support epithelial differentiation, nutrient transport, barrier formation, and interactions with the microbiota under defined conditions. In vitro intestine models help researchers investigate digestion, absorption, inflammation, infection, and drug toxicity while reducing reliance on animal studies. By linking cellular mechanisms to tissue-level function, they also support advances in personalized medicine, regenerative biology, and the development of more predictive therapies.

Performing cell culture assays for investigating bacterial adhesion and invasion under aerobic conditions is usually unrepresentative of the in vivo environment. A Vertical Diffusion Chamber model allows study of interactions of the human pathogen Campylobacter jejuni with intestinal epithelial cells under more in vivo-like conditions, resulting in enhanced bacterial invasion.

In this video, we demonstrate a technique to isolate whole crypts from a mouse’s small intestine. The isolated whole crypts can be used to propagate long-lived and self-renewing 3D organoids in vitro.

We describe a novel surgical method for catheterizing 'intestinal loops' within the ileum of sheep. Once animals have recovered from surgery and have cleared antibiotics and analgesics, multiple treatments can be deposited directly in loops via the catheters.
