Estrogen Receptor Alpha

Estrogen receptor alpha (ERα) is a ligand-activated nuclear receptor that regulates gene expression in response to estrogen, making it a key mediator of reproductive biology, development, and tissue function. After estrogen binds ERα, the receptor undergoes a conformational change, forms dimers, and binds estrogen response elements in DNA or interacts with other transcriptional regulators to control target genes. Studying ERα helps explain hormone-dependent processes in tissues such as the breast, uterus, and bone, while assays that measure its expression or activity support research on endocrine signaling, breast cancer biology, and therapies that block or modify estrogen-responsive pathways.

Estrogen Receptor-Reporter Activity Assay to Screen Phytoestrogen Estrogenic Activity

This video describes a cell-based reporter assay to determine the estrogenic activity of plant-derived compounds. A compound with estrogen-like activity mimics its binding to estrogen receptors and drives downstream reporter enzyme gene transcription. The reporter enzyme generates bioluminescence in a reaction, indicating the estrogenic activity of the compound.

Molecular signaling through both estrogen and microRNAs are critical in breast cancer development and growth. Estrogen activates the estrogen receptors, which are transcription factors. Many transcription factors can regulate the expression of microRNAs, and estrogen-regulated microRNAs can be profiled using different large-scale techniques.
