Fibroblast Subpopulations

Fibroblast subpopulations are distinct groups of fibroblasts within connective tissues that differ in molecular markers, developmental history, and biological function, making them important for understanding tissue organization and disease. Their identities and behaviors arise from interactions between intrinsic gene-regulatory programs and local signals, including transforming growth factor beta, inflammatory cytokines, and mechanical cues that control extracellular-matrix production, remodeling, and contractility. Researchers use approaches such as single-cell RNA sequencing, spatial profiling, and lineage tracing to characterize these populations in health and injury. This knowledge helps clarify wound repair, organ fibrosis, tumor-stroma interactions, and strategies for tissue engineering and regenerative medicine.

Leukemic Subpopulation Harvest: A Method for Spatial Separation of Leukemic Cell Subpopulations from 2D Co-culture

2023

This video describes a method for spatial separation of three leukemic cell subpopulations: the suspended leukemic cells freely floating in the media, phase bright leukemic cells adhered to the surface of the mesenchymal stromal cell monolayer, and the phase dim leukemic cells that have migrated beneath the mesenchymal stromal cell monolayer from 2D co-culture.

Fibroblast behavior underlies a spectrum of clinical entities, but they remain poorly characterized, largely due to their inherent heterogeneity. Traditional fibroblast research relies upon in vitro manipulation, masking in vivo fibroblast behavior. We describe a FACS-based protocol for the isolation of mouse skin fibroblasts that does not require cell culture.

Purification of Subpopulations of Extracellular Vesicles: A Technique to Purify Subpopulations of Extracellular Vesicles Using Density Gradient Ultracentrifugation

Carcinoma-associated fibroblasts (CAFs) rich in myofibroblasts present within the tumour stroma, play a major role in driving tumour progression. We developed a coimplantation tumour xengraft model for experimentally generating CAFs from human mammary fibroblasts. The protocol describes how to establish CAF myofibroblasts that acquire an ability to promote tumourigenesis.

Flow Cytometry Staining of Mycobacterium tuberculosis Infected Macrophage Subpopulations

This video showcases a flow cytometric gating strategy utilizing a multi-color panel to analyze cell viability and specific fluorescent-conjugated anti-human antibody markers. This approach makes it possible to determine the percentage of Mycobacterium tuberculosis-infected cell populations accurately.
