Fluorescent Reporter

A fluorescent reporter is a molecule or genetically encoded protein that produces a detectable fluorescent signal, enabling researchers to visualize biological activity in living cells and tissues. In biology, a reporter gene such as green fluorescent protein is placed under a chosen promoter or fused to a protein of interest; when expressed, its fluorophore absorbs excitation light and emits light at a longer wavelength. Fluorescent reporters help measure gene expression, track protein localization, monitor cell signaling, and follow developmental or disease-related processes over time. Their compatibility with microscopy and quantitative imaging makes them valuable for studying dynamic cellular mechanisms and evaluating experimental treatments.

This video demonstrates the reporter enzyme fluorescence (REF) imaging for in vivo detection of Mycobacterium tuberculosis infection. The infected mice are intraperitoneally injected with a substrate for REF imaging. Upon being hydrolyzed by bacterial β-lactamase enzyme, the substrate emits fluorescence, the detection of which aids evaluation of infection.
