Fluorescent Spot Detection

Fluorescent spot detection is an image-analysis method that identifies and localizes small, bright fluorescent features in microscopy images, enabling researchers to study molecules, organelles, cells, and dynamic biological events. The method distinguishes spots from background by evaluating pixel intensity, applying thresholding or filtering, and often segmenting each feature to measure its position, size, brightness, or movement. In biology, fluorescent spot detection supports the analysis of protein localization, vesicle trafficking, chromosome behavior, and single-molecule dynamics. Reliable detection can reveal changes in molecular distribution and interactions over time, while automated workflows improve measurement consistency across large image datasets.

This video demonstrates a method to detect NLRP3 inflammasome activation in macrophages using fluorescent reporter probes. The cells are treated with potassium ionophores and glycine to induce inflammasome assembly and caspase-1 activation. Fluorescent reporter probes visualize activated caspase-1, and cells are stained with a fluorescent dye for nuclei visualization.
