Human X3p Genechip

The Human X3P GeneChip is a microarray platform for measuring the expression of thousands of human genes simultaneously, supporting large-scale studies of cellular function and regulation. It works by hybridizing fluorescently labeled RNA-derived targets to complementary oligonucleotide probes arranged on a solid surface; the resulting signal intensity reflects the relative abundance of corresponding transcripts. Researchers use this gene expression profiling approach to compare biological samples, identify differentially expressed genes, characterize disease-associated molecular changes, and investigate responses to experimental conditions. By converting transcript abundance into a genome-wide expression profile, the platform helps connect molecular activity with biological phenotypes.

In Vivo Modeling of the Morbid Human Genome using Danio rerio

Here, we present a systematic approach for developing physiologically relevant, sensitive and specific in vivo assays for interpreting variation in human pathology. Transient genetic manipulation via microinjection of WT and mutant human mRNA and morpholino (MO) antisense oligonucleotides harness the tractability of the developing zebrafish embryo to rapidly assay pathogenic mutations, especially, but not exclusively, in the context of human developmental disorders.

Techniques for Imaging Ca2+ Signaling in Human Sperm

Stimulus-evoked [Ca2+]i signals of individual human sperm are assessed. Motile cells are loaded with Ca2+-sensitive fluorescent dye (AM-ester method) and immobilised in a perfusable chamber. Cells are imaged by time-lapse fluorescence microscopy and stimulated via the perfusing medium. Responses of single cells (or regions) are analysed offline using Excel.

Lentiviruses are a valuable research tool for exploring gene function; however, researchers may wish to avoid production of pantropic lentivirus encoding known or suspected oncogenes. As an alternative, we present a safer protocol for use of ecotropic lentivirus on human cells modified to express the ecotropic receptor mSlc7a1.
