Fragment Ligated Inhibitory Peptides

Fragment ligated inhibitory peptides are engineered peptide inhibitors assembled by joining shorter peptide fragments into a functional sequence, providing a way to target specific biological interactions. In a typical design, separately prepared fragments are chemically ligated to reconstruct the binding region; the resulting peptide recognizes a target protein and interferes with an active site, interaction surface, or signaling event. In biology, these molecules help test protein function, identify interaction interfaces, and dissect regulatory pathways. Their modular construction can also support optimization of peptide stability, specificity, and activity, making fragment ligation useful for developing experimental probes and potential therapeutic candidates.

DNA Ligation Reactions

2023

In molecular biology, ligation refers to the joining of two DNA fragments through the formation of a phosphodiester bond. An enzyme known as a ligase catalyzes the ligation reaction. In the cell, ligases repair single and double strand breaks that occur during DNA replication. In the laboratory, DNA ligase is used during molecular cloning to join DNA fragments of inserts with vectors – carrier DNA molecules that will replicate target fragments in host organisms. This protocol describes a method for producing and evaluating the antimicrobial activity and cytotoxicity of the cationic peptide Mel4 and peptide mimic RK758. By integrating chemical synthesis, recombinant expression, and standardized functional assays, it provides a streamlined workflow for the production and testing of peptides and small-molecule mimics.

Habitat Fragmentation

2020

Habitat fragmentation describes the division of a more extensive, continuous habitat into smaller, discontinuous areas. Human activities such as land conversion, as well as slower geological processes leading to changes in the physical environment, are the two leading causes of habitat fragmentation. The fragmentation process typically follows the same steps: perforation, dissection, fragmentation, shrinkage, and attrition. Perforation and dissection often occur during the initial stages of...
