Fungal Cell Wall

The fungal cell wall is a dynamic extracellular layer that surrounds fungal cells, providing structural support, protection, and control over cell shape. Built primarily from chitin, β-glucans, and mannoproteins, it forms a fibrous network that resists internal osmotic pressure while being continuously remodeled during growth, budding, and hyphal extension. Its distinctive composition helps fungi survive changing environments and makes the wall a major focus in biology and medicine. Research on cell-wall assembly supports fungal classification, studies of host-pathogen interactions, and development of antifungal drugs that disrupt wall synthesis without targeting the structurally different cells of humans.

We describe methods for live-cell video microscopy of Candida albicans phagocytosis by macrophages. These methods enable stage-specific analysis of macrophage migration, recognition, engulfment and phagosome maturation and reveal novel aspects of phagocytosis.

Glycan Profiling of Plant Cell Wall Polymers using Microarrays

A technique called Comprehensive Microarray Polymer Profiling (CoMPP) for the characterisation of plant cell wall glycans is described. This method combines the specificity of monoclonal antibodies directed to defined glycan-epitopes with a miniature microarray analytical platform allowing screening of glycan occurrence in a broad range of biological contexts.
