Hypoxia Induction

Hypoxia induction is the deliberate creation of low-oxygen conditions in cells, tissues, or organisms to study how biology responds to oxygen limitation. Reduced oxygen availability inhibits oxygen-dependent prolyl hydroxylation of hypoxia-inducible factors, allowing HIF proteins to accumulate, enter the nucleus, and activate genes involved in metabolism, angiogenesis, and survival. Researchers induce hypoxia by controlling oxygen levels in culture systems or by using chemical agents that mimic oxygen deprivation. This approach helps investigate developmental adaptation, tumor biology, ischemic injury, inflammation, and cellular energy regulation, while supporting the evaluation of therapies that target oxygen-sensing pathways.

Induction of Hypoxia in Living Frog and Zebrafish Embryos

We introduce a novel hypoxic chamber system for use with aquatic organisms such as frog and zebrafish embryos. Our system is simple, robust, cost-effective and allows the induction and sustainment of hypoxia in vivo and for up to 48 h. We present 2 reproducible methods to monitor the effectiveness of hypoxia.

Induction and Characterization of Pulmonary Hypertension in Mice using the Hypoxia/SU5416 Model

This protocol describes the induction of pulmonary hypertension (PH) in mice based on the exposure to hypoxia and the injection of a VEGF receptor antagonist. The animals develop PH and right ventricular (RV) hypertrophy 3 weeks after the initiation of the protocol. The functional and morphometrical characterization of the model is also presented.

Thromboembolic stroke models are vital tools for optimizing the recanalization therapy. Here we report a murine thrombotic stroke model based on transient cerebral hypoxic-ischemic (tHI) insult, which triggers thrombosis and infarction, and responds favorably to tissue plasminogen activator (tPA)-mediated fibrinolysis in a therapeutic window similar to those in stroke patients.
