Larval Zebrafish Dissection

Larval zebrafish dissection is a microsurgical method for opening and examining zebrafish larvae to study their developing organs, tissues, and anatomical organization. Researchers typically anesthetize larvae, position them under a stereomicroscope, and use fine instruments to remove or separate selected tissues while preserving structures for imaging, staining, or molecular analysis. Because larval zebrafish are small and develop rapidly, dissection supports investigations of embryonic development, organ formation, regeneration, and disease models. The technique connects whole-animal biology with cellular and tissue-level observations, helping researchers relate visible anatomy to developmental and physiological processes.

A simple method to record extracellular field potentials in the larval zebrafish forebrain is described. The method provides a robust in vivo read-out of seizure-like activity. This technique can be used with genetically modified zebrafish larvae carrying epilepsy-related genes or seizures evoked by administration of convulsant drugs.

Source: Li, J., et al. Development of a Larval Zebrafish Infection Model for Clostridioides difficile. J. Vis. Exp. (2020).This video demonstrates the microgavage procedure, where an anesthetized zebrafish larva is immobilized in agarose and orally injected with a pathogenic bacterium using a tracer-guided needle. The non-invasive delivery mimics natural infection, enabling analysis of bacterial replication, toxin release, and intestinal epithelial damage.
