Microinjector Use

Microinjector use is the controlled delivery of small volumes of fluid into cells, embryos, or other microscopic biological structures, enabling precise manipulation that bulk methods cannot provide. A fine glass micropipette, positioned with a micromanipulator, penetrates the target membrane while applied pressure or mechanical displacement expels a measured volume of material. In biology, microinjection supports gene delivery, embryo manipulation, intracellular labeling, and the study of cell function by introducing DNA, RNA, proteins, dyes, or other reagents directly into defined compartments. Its precision makes it valuable for developmental biology, transgenesis, cell-based research, and experimental models of disease.

2025

Source: Poland, R. et al. Rodent Brain Microinjection to Study Molecular Substrates of Motivated Behavior. J. Vis. Exp. (2015)In this video, a water-filled microinjection setup consisting of a syringe and microinjector tubing is used for precise drug delivery. A rat with a pre-implanted guide cannula undergoes targeted brain microinjection, enabling accurate administration of therapeutic agents in awake animals with minimal tissue damage.

(2020)This video demonstrates the protocol for microinjection of tetrodotoxin (TTX) into the hypothalamus of a freely moving rat to transiently inhibit neuronal activity, suppress gonadotropin-releasing hormone secretion, and prevent ovulation.

Microinjection of Drosophila Nurse Cells: A Method of Intracellular Delivery

2023

Due to their large size and relatively simple organization, Drosophila nurse cells are ideally suited for live cell imaging studies. This video describes a microinjection method for exogenous compounds delivery into the cells, and the featured protocol demonstrates the procedure with fluorescent oligonucleotide probes called molecular beacons (MBs) used to visualize endogenous mRNA transcripts.

Bacterial Endophthalmitis Mouse Model: A Method for Generating Bacterial Endophthalmitis via Intravitreal Injection of Bacterial Suspension into Eye of Mouse

2025

This video describes a method for generating a mouse model that mimics bacterial endophthalmitis – an intraocular infection resulting from a bacterial invasion into the eye’s vitreous cavity – via intravitreal injection of a bacterial suspension into the eye. This mouse model helps study the disease mechanism and evaluate therapeutics for it.

Zebrafish Microinjection Techniques

2023

One of the major advantages to working with zebrafish (Danio rerio) is that their genetics can be easily manipulated by microinjection of early stage embryos. Using this technique, solutions containing genetic material or knockdown constructs are delivered into the blastomeres: the embryonic cells sitting atop the yolk of the newly fertilized egg. Delivery into the cytoplasm is achieved either through direct injection into the blastomere, or via natural cytoplasmic movements that occur after a...
