Microvolume Spectrophotometer

A microvolume spectrophotometer is an analytical instrument that measures the absorbance of microliter-scale biological samples, enabling rapid analysis while conserving valuable material. It uses a short optical path and UV-visible light to determine how strongly a sample absorbs specific wavelengths; the Beer-Lambert law relates absorbance to the concentration of molecules such as nucleic acids and proteins. In biology, researchers use these measurements to quantify DNA, RNA, and protein preparations, assess sample purity, and identify potential contamination before downstream experiments such as PCR, sequencing, or protein analysis. This approach supports efficient, low-waste workflows when sample volume is limited.

2023

The spectrophotometer is a routinely used instrument in scientific research. Spectrophotometry is the quantitative measurement of how much a chemical substance absorbs light by passing a beam of light through the sample using a spectrophotometer. In this video, basic concepts in spectrophotometry, including transmittance, absorbance and the Beer-Lambert Law are reviewed in addition to the components of the spectrophotometer. These concepts provide a foundation for how to determine the...
