Nadh2 Production Measurement

NADH2 production measurement quantifies the formation of reduced nicotinamide adenine dinucleotide, a key electron carrier in cellular energy metabolism. The assay commonly tracks changes in NADH2 concentration through its characteristic absorbance at 340 nm, using increases in optical density to indicate cofactor production or enzyme activity under defined reaction conditions. In biology, these measurements help assess cellular respiration, redox balance, metabolic pathway activity, and the function of NADH-generating enzymes. They also support studies of mitochondrial performance, bioenergetics, and metabolic disorders by linking cofactor production to biochemical and cellular outcomes.

Glycolysis is a defining metabolic marker in multiple biological systems. Monitoring glycolysis by measuring the extracellular flux of H+ is common, but requires correction to be quantitative and unambiguous. Here, we demonstrate how to gather and correct extracellular flux data to distinguish between respiratory and glycolytic sources of extracellular acidification.
