Neutrophil Migration Analysis

Neutrophil Migration Analysis is the study of how neutrophils move through tissues toward sites of infection, injury, or inflammation, providing insight into innate immune defense. The process begins when chemical signals such as chemokines and bacterial peptides create gradients that neutrophils detect through surface receptors; cells then polarize, reorganize actin, adhere to endothelial cells, and migrate by directed crawling and tissue penetration. Researchers analyze these behaviors using live-cell imaging, wound-healing assays, and transwell or microfluidic chemotaxis systems. The results help clarify inflammatory mechanisms, evaluate immune-cell function, and identify potential targets for treating infection, chronic inflammation, and immune-related disease.

Quantitative Assessment of Human Neutrophil Migration Across a Cultured Bladder Epithelium

We developed an in vitro model that mimics an important component of the acute inflammatory response during infection of the bladder with uropathogenic Escherichia coli. The transuroepithelial neutrophil migration assay enables quantitative assessment of human neutrophil migration across bladder epithelia, cultured on permeable supports, in response to bacterial infection or chemoattractant substances.

We describe a protocol of brightfield intravital microscopy for measuring dynamic neutrophil-endothelial cell interactions during neutrophil recruitment in response to the source of a neutrophil chemoattractant in vivo. Neutrophil intraluminal crawling, transendothelial migration and chemotaxis in mouse cremaster muscle tissue are visualized with time-lapsed video photography and tracked with ImageJ.
