Non Canonical Pathways

Non-canonical pathways are alternative biological routes that operate outside, or alongside, the best-characterized canonical mechanisms, expanding how cells process signals and regulate responses. They may use different receptors, adaptor proteins, enzymes, or downstream effectors, and are often activated by distinct cellular conditions, allowing overlapping signals to produce context-dependent outcomes. In biology, studying these pathways clarifies how cells adapt to stress, communicate across tissues, and maintain homeostasis when standard routes are altered. Their analysis also helps explain disease mechanisms and supports the development of more precise experimental strategies and therapeutic targets.

Non-Canonical Wnt Signaling Pathways

2023

Wnt is a zygotic effect gene that is expressed during very early embryonic development. It regulates various processes in animals starting from early development through the adult stage, such as organogenesis in the embryo and maintenance of neuronal and blood stem cells. Wnt proteins can induce a wide variety of intracellular pathways depending upon the specific abilities of different Wnt ligands to form a complex with shared and cognate receptors in the presence of different co-receptors. The...

The gene encoding the main signaling molecules of the Wnt signaling pathways (the Wnt proteins) was discovered almost four decades ago by Nüsslein-Volhard and Wieschaus. They identified and originally named the gene "wingless" (wg) after a phenotype discovered during their landmark genetic screen in Drosophila for body pattern defects. At around the same time, another researcher named Harold Varmus found that a murine tumor virus activates the mammalian wg homolog, Int-1, which results in tumor...

The protocol presents the Escherichia coli-based selective pressure incorporation of non-canonical amino acids (ncAAs) into the lactococcal antimicrobial peptide nisin. Its properties can be changed during recombinant expression via substitution with desired ncAAs in defined growth media. Resulting changes in bioactivity are mapped by growth inhibition assays and fluorescence microscopy.
