Ovary Wall

The ovary wall is the tissue that surrounds and protects ovules within the flower’s ovary, and it becomes the fruit wall in flowering plants. After fertilization, signaling associated with seed and embryo development stimulates ovary-wall growth and differentiation into the pericarp, commonly organized into outer exocarp, middle mesocarp, and inner endocarp layers. These tissues can form protective shells, fleshy edible tissues, or dispersal structures, depending on the species. Studying the ovary wall helps explain fruit development, seed protection, plant reproduction, and the traits that influence crop quality, yield, storage, and breeding.

A technique called Comprehensive Microarray Polymer Profiling (CoMPP) for the characterisation of plant cell wall glycans is described. This method combines the specificity of monoclonal antibodies directed to defined glycan-epitopes with a miniature microarray analytical platform allowing screening of glycan occurrence in a broad range of biological contexts.

How niches and stem cells form during development is an important question with practical implications. In the Drosophila ovary, germ line stem cells and their somatic niches form during larval development. This video demonstrates how to dissect, stain and mount female gonads from late third instar (LL3) Drosophila larvae.

This protocol describes the primary culture/co-culture of mouse ovarian tissue, using ovaries from neonatal mice and individual ovarian follicles from prepubertal mice. The culture techniques support development in a highly physiological manner, allowing investigation of the effect of extrinsic agents on the ovary, and of interactions between ovarian follicles.
