Parafilm Method

The Parafilm Method is a laboratory technique that uses flexible, self-sealing paraffin film to close and protect tubes, flasks, plates, and other containers during biological experiments. When stretched around a vessel opening, the film conforms to the rim and forms a temporary barrier that helps limit contamination, moisture loss, and exposure to airborne particles while accommodating routine handling. In biology, researchers use this method to support cell and microbial cultures, preserve biological samples, and secure containers during incubation, transport, or storage. Because it is quick, inexpensive, and removable, Parafilm provides a practical supplement to standard laboratory closures.

Stress Granules (SGs) are nonmembranous cytoplasmic structures that form in cells exposed to a variety of stresses. SGs contain mRNAs, RNA-binding proteins, small ribosomal subunits, translation-related factors, and various cell signaling proteins. This protocol describes a workflow that uses several experimental approaches to detect, characterize, and quantify bona fide SGs.
