Parkinson's Disease Model

A Parkinson’s disease model is an experimental system that reproduces selected features of Parkinson’s disease to investigate its causes, progression, and potential treatments. Models may use cultured neurons, patient-derived cells, organoids, or animals to examine mechanisms such as dopaminergic neuron loss, α-synuclein accumulation, mitochondrial dysfunction, and neuroinflammation. In biology research, these systems help scientists connect cellular and molecular changes with motor symptoms, assess biomarkers, and test drug candidates or gene-based interventions. Because no single model captures the full human condition, comparing complementary models improves understanding of disease mechanisms and supports the development of more predictive therapies.

Murine Model for Parkinson's Disease: from 6-OH Dopamine Lesion to Behavioral Test

Parkinson disease is caused by loss of dopaminergic innervation to the striatum, which can be experimentally induced by 6-OH-dopamine. We describe how to perform a stereotaxic lesion and to monitor apomorphine-induced rotational behavior in mice. This model is useful and reliable for testing new therapies for Parkinson disease.

Histological Examination of Mitochondrial Morphology in a Parkinson's Disease Model

This study presents a method to analyze the morphology of mitochondria based on immunostaining and image analysis in mouse brain tissue in situ. It also describes how this allows one to detect changes in mitochondrial morphology induced by protein aggregation in Parkinson's disease models.

In Parkinson's disease and movement disorders in general, sensitive and reliable behavioral assays are essential for testing novel potential therapeutics. Here, we describe a manageable battery of sensorimotor tests for mice that are sensitive to varying degrees of injury to the nigrostriatal system and useful for preclinical studies.
