Peripheral Blood Culture

Peripheral blood culture is a microbiological method that detects microorganisms circulating in the bloodstream, making it essential for investigating suspected bloodstream infections. Blood collected by peripheral venipuncture is introduced into sterile aerobic and anaerobic culture bottles, where nutrient broth supports microbial growth; automated systems monitor changes such as carbon dioxide production and signal when growth occurs, followed by organism identification and antimicrobial susceptibility testing. In biology and clinical microbiology, peripheral blood culture helps diagnose bacteremia and sepsis, guide targeted treatment, and distinguish true infection from contamination. Proper collection technique and skin antisepsis are critical for reliable results.

Generation and Culture of Blood Outgrowth Endothelial Cells from Human Peripheral Blood

This protocol allows for the reliable generation and characterization of blood outgrowth endothelial cells (BOECs) from a small volume of adult peripheral blood. BOECs can be used as a surrogate for endothelial cells from patients with vascular disorders and as a substrate for the generation of induced pluripotent stem cells.

In this video, we describe a protocol for human primary T cell activation. The incubation of naïve T cells with magnetic beads coated with anti-CD3 and anti-CD28 antibodies, followed by interleukin-2 or interleukin-15 treatment, leads to the activation of naïve T cells and their differentiation and proliferation into effector or memory T cells.

A method to expand γδ T cells from peripheral blood mononuclear cells (PBMC) is described. PBMC-derived γδ T cells are stimulated and expanded using zoledronate and interleukin-2 (IL-2). Large scale expansion of γδ T cells can be applied to autologous cellular immunotherapy of cancer.
