Phenotypic Rescue Assay

A phenotypic rescue assay is an experimental method that tests whether restoring a gene, protein, or pathway can reverse an abnormal phenotype caused by a specific perturbation. Researchers first create or identify a phenotype through gene knockdown, mutation, inhibition, or environmental manipulation, then reintroduce the functional component or apply a candidate treatment and compare the outcome with appropriate controls. Recovery of the phenotype supports a causal link between the disrupted factor and the observed biological effect, although incomplete rescue can indicate dosage, timing, or pathway complexity. In biology, these assays help validate gene function, clarify molecular mechanisms, and evaluate potential therapeutic strategies.

A parasite-rescue and transformation assay with THP1 cells infected in vitro with Leishmania donovani has been optimized for anti-leishmanial drug screening. The assay involves differentiation of THP1 cells, infection with promastigotes, treatment with test drugs, controlled lysis of the infected macrophages, rescue of amastigotes, transformation to promastigotes and monitoring promastigote growth and proliferation with a fluorometric assay.
