Lung Endothelium

Lung endothelium is the specialized layer of cells lining the pulmonary blood vessels, where it regulates communication between circulating blood and lung tissue. These endothelial cells form a selective barrier that controls fluid and solute movement, supports gas exchange, and responds to signals through cell adhesion, vascular permeability, and inflammatory pathways. The lung endothelium also helps coordinate blood flow across the pulmonary circulation and interacts with immune cells, platelets, and airway tissues. Studying its structure and function is essential for understanding pulmonary inflammation, edema, thrombosis, vascular remodeling, and diseases that impair respiratory and cardiovascular health.

This is a method to visualise leukocyte adhesion to the endothelium in harvested pressurised vessels. The technique enables studying vascular adhesion under shear flow with differing intraluminal pressures up to 200 mmHg thus mimic-ing the pathophysiological conditions of high blood pressure.

After puncturing the aorta through the inferior vena cava (IVC) to create an aorto-caval fistula in the mouse, solution containing a drug is infused into the IVC via the same needle, followed by incubation. This method enables more robust drug delivery to the venous endothelium compared to the external route.

We have developed a decellularized lung extracellular matrix and novel biomimetic bioreactor that can be used to generate functional lung tissue. By seeding cells into the matrix and culturing in the bioreactor, we generate tissue that demonstrates effective gas exchange when transplanted in vivo for short periods of time.

This paper describes a striaghforward and efficient method of intubating mice for pulmonary function measurements or pulmonary instillation, that allows the mice to recover and be studied at later times. The procedure involves an inexpensive fiberoptic light source that directly illuminates the trachea.

Lung Capacity

2019

The air in the lungs is measured in volumes and capacities. Lung volume measures reflect the amount of air taken in, released, or left over after a lung function, like a single inhalation. Lung capacity measures are sums of two or more lung volume measures. There are four defined lung volume measures: tidal volume, inspiratory reserve volume, expiratory reserve volume, and residual volume. Tidal volume is the amount of air inhaled and exhaled in a normal breath. Inspiratory...
