Pick And Plunge Mechanism

The pick-and-plunge mechanism is a cryogenic sample-preparation approach that transfers a specimen-coated electron microscopy grid into a cryogen for rapid freezing, preserving biological material in a near-native state. A mechanical arm or holder picks up the grid, positions it over liquid ethane or another suitable cryogen, and plunges it quickly so the thin aqueous film vitrifies rather than forming damaging ice crystals. This controlled motion supports preparation of cryo-electron microscopy specimens for imaging proteins, viruses, membranes, and other cellular structures. Consistent vitrification improves structural preservation and can increase the quality and interpretability of high-resolution biological reconstructions.
