Plasmid Amplification

Plasmid amplification is the production of many copies of a circular, extrachromosomal DNA molecule, typically in a host bacterium, to obtain sufficient material for biological research. The process begins when a plasmid enters competent cells by transformation; antibiotic selection enriches cells carrying the plasmid, and growth in culture allows host replication machinery to duplicate its origin-directed DNA as cells divide. Researchers then harvest the culture and purify the plasmid through cell lysis, removal of cellular contaminants, and DNA recovery. Amplified plasmids support cloning, gene expression, sequencing, mutagenesis, and recombinant protein studies, making the method a foundation of molecular biology.

Plasmid purification is a technique used to isolate and purify plasmid DNA from genomic DNA, proteins, ribosomes, and the bacterial cell wall. A plasmid is a small, circular, double-stranded DNA that is used as a carrier of specific DNA molecules. When introduced into a host organism via transformation, a plasmid will be replicated, creating numerous copies of the DNA fragment under study.
