Preimplantation Development

Preimplantation development is the early sequence of embryonic changes from fertilization to implantation in the uterine lining, making it a critical transition in mammalian reproduction. After sperm-egg fusion, the zygote undergoes repeated cleavage divisions, compacts into a morula, and forms a blastocyst with distinct inner cell mass and trophectoderm layers. The blastocyst must maintain developmental potential while traveling through the oviduct and coordinating implantation with the uterus. Studying this process clarifies early cell fate decisions, embryonic genome activation, and causes of developmental failure, while supporting assisted reproductive technologies, embryo assessment, and research on fertility and early pregnancy.

Quantitative Analysis of Protein Expression to Study Lineage Specification in Mouse Preimplantation Embryos

This protocol presents a method to perform quantitative, single-cell in situ analysis of protein expression to study lineage specification in mouse preimplantation embryos. The procedures necessary for collection of blastocysts, whole-mount immunofluorescent detection of proteins, imaging of samples on a confocal microscope, and nuclear segmentation and image analysis are described.

Cryopreservation of Preimplantation Embryos of Cattle, Sheep, and Goats

Preimplantation embryos may be cryopreserved after placement into a hypertonic cryoprotective solution to cause cellular dehydration. After equilibration, ice crystal formation is induced in the solution surrounding the embryo. Further dehydration occurs as the embryo is slowly cooled to subzero temperatures before plunging into liquid nitrogen for storage.
