Porcine Ischemic Model

Porcine ischemic models are experimental animal systems that reproduce tissue injury caused by inadequate blood flow, providing a biologically relevant platform for studying ischemia in vivo. Researchers create a controlled reduction or interruption of perfusion in pigs, producing oxygen and nutrient deprivation that can lead to cellular dysfunction, inflammation, and organ damage; restoring circulation can then model reperfusion injury when included in the design. These models support investigation of ischemic pathophysiology, tissue responses, imaging and biomarker development, and the safety and efficacy of therapeutic interventions. Their physiological scale also strengthens translation of findings from laboratory studies toward clinical research.

The rat, due to its size, availability, and rather docile behavior, has been utilized as a research model for many years. The goal of this protocol is to utilize the rat as an ischemic skin wound healing model to provide valuable insight into the pathophysiology of chronic wounds.

A Simplified Technique for Producing an Ischemic Wound Model

We have developed a minimally invasive technique to create a rabbit ischemic ear wound model by dividing the central artery and nerve and the cranial neurovascular bundle. A subcutaneous tunnel then cuts all subcutaneous tissues. This procedure causes minimal skin disruption and can be safely used in diabetic animals.
