Proofreading Mechanism

Proofreading mechanisms are biological fidelity checks that reduce errors during DNA replication and other nucleic acid synthesis processes, helping preserve accurate genetic information. During replication, a DNA polymerase selects a complementary nucleotide, detects a mismatch through structural and chemical interactions, and transfers the newly formed 3′ end to an exonuclease site, where the incorrect nucleotide is removed before synthesis resumes. This mechanism greatly lowers replication error rates and supports genome stability, while defects can increase mutation frequency. Understanding proofreading is important for studying heredity, disease-associated mutations, genome evolution, and the design of biochemical and molecular biology methods.
