Rat Paw Model

The rat paw model is an in vivo experimental system used to study inflammation, tissue swelling, pain, and responses to therapeutic compounds. Researchers commonly inject an inflammatory agent, such as carrageenan, into the paw, triggering mediator release, increased vascular permeability, and fluid accumulation that produce measurable edema and sensitivity. Paw thickness or volume, along with thermal or mechanical pain behaviors, provides quantitative endpoints for evaluating inflammatory and analgesic effects. In biology and pharmacology, this model supports investigation of inflammatory mechanisms, comparison of candidate treatments, and assessment of drug efficacy before more advanced preclinical studies.

Classical forelimb asymmetry analysis of the cylinder test is routinely used to assess behavioural deficits in rats following brain injury or stroke; however, it fails to detect consistent deficits in mice. This study demonstrates that quantifying paw-dragging behaviour is a more sensitive analysis of brain injury in mice.

Source: Brakkee, E. M., et al. Surgery and Behavioral Testing in the Tibial Neuroma Transposition Model in Rats. J. Vis. Exp. (2023).This video demonstrates a procedure to generate a rat tibial neuroma transposition (TNT) model. After exposing the tibial nerve in the hind leg of an anesthetized rat, transect the nerve. Next, create a subcutaneous tunnel, thread the severed nerve through the tunnel, and suture it in place. In the absence of a target for reinnervation, disorganized axonal...
