Rbp2 Isoform

RBP2 isoforms are related protein variants produced from the RBP2, also known as KDM5A or JARID1A, gene, and they can differ in structure, expression, or regulatory activity. RBP2 functions as a chromatin regulator and histone demethylase, using an iron- and 2-oxoglutarate-dependent catalytic mechanism to remove methyl groups from histone H3 lysine 4, while protein domains support interactions with transcriptional regulators such as the retinoblastoma protein. Studying RBP2 isoforms helps clarify how alternative protein forms influence gene expression, cell-cycle control, development, and disease-associated changes in chromatin, including mechanisms relevant to cancer biology.

Here we are presenting a chromatin immunoprecipitation (ChIP) procedure for genome-wide location analysis of protein isoforms that differ in a histone-binding domain. We are applying it to ChIP-Seq analysis to identify the targets of the KDM5A/JARID1A/RBP2 histone demethylase.

We describe a high-throughput, multiplex, and targeted proteomic cerebrospinal fluid (CSF) assay developed with potential for clinical translation. The test can quantitate potential markers and risk factors for neurodegeneration, such as the apolipoprotein E variants (E2, E3 and E4), and measure their allelic expression.
