Swab Scraping Technique

The swab scraping technique is a biological sampling method that collects microorganisms, cells, or surface material by rubbing and lightly scraping a defined area with a sterile swab. Mechanical friction dislodges material from the surface, which is then transferred to a culture medium, slide, or extraction buffer for analysis. Researchers use this technique in microbiology, environmental sampling, clinical testing, and laboratory investigations of surface-associated communities. Consistent sampling area, pressure, timing, and aseptic handling improve reproducibility, while subsequent culturing, microscopy, or molecular assays can reveal microbial presence, cellular features, or contamination levels.

A citizen science project was designed to recruit San Diego residents to collect environmental samples for SARS-CoV-2. A multilingual web-based platform was created for data submission using a user-friendly mobile device interface.
