Reporter Assay

A reporter assay is a biological measurement method that uses an observable or quantifiable signal to reveal the activity of a specific gene, promoter, enhancer, or signaling pathway. It works by linking a regulatory sequence or molecular response element to a reporter gene, such as luciferase or green fluorescent protein, so that changes in transcription produce a measurable light or fluorescence signal. Researchers use reporter assays to study gene regulation, transcription factor activity, cellular signaling, and responses to drugs or other experimental conditions. By converting complex molecular events into quantitative readouts, these assays support mechanistic studies and help evaluate regulatory elements or candidate therapeutics.

Massively Parallel Reporter Assays in Cultured Mammalian Cells

The genetic reporter assay is a well-established and powerful tool for dissecting the relationship between DNA sequences and their gene regulatory activities. Coupling candidate regulatory elements to reporter genes that carry identifying sequence tags enables massive parallelization of these assays.
