Semen Collection Technique

Semen collection technique is a controlled biological method for obtaining ejaculate so its cellular and biochemical properties can be examined or used in reproductive research. Collection typically stimulates ejaculation through an appropriate method for the species and setting, while a sterile, labeled container protects the sample from contamination and preserves its quality for analysis. Researchers and clinicians can then evaluate volume, sperm concentration, motility, morphology, and viability, making the technique important for fertility assessment, semen preservation, artificial insemination, assisted reproduction, and studies of male reproductive biology.

2025

This protocol presents a simple microaspiration technique to isolate viable somatic cells (SCs) from cryopreserved equine semen. Unlike traditional approaches, this technique avoids long in vitro cultures and overcomes sperm contamination, enabling immediate use in somatic cell cloning.

Collection of Post-mating Semen from the Female Reproductive Tract and Measurement of Semen Liquefaction in Mice

Sperm Structure and Semen Composition

2024

During ejaculation, males release around 2-5 milliliters of semen, which is a complex mixture of mature sperm and various fluids produced by accessory glands. The mature sperm cells measure approximately 60 micrometers in length and consist of a head, neck, midpiece, and tail. The head is flattened and tapered, measuring about 4 to 5 micrometers in length. It contains a nucleus with condensed chromosomes and an acrosome, a cap-like structure filled with enzymes essential for penetrating the...

Technique to Collect Fungiform (Taste) Papillae from Human Tongue

Knowledge of molecular mechanisms underlying gustatory transduction has recently enjoyed significant advances, largely due to using animal models. However, the wide diversity in taste sensitivity and specificity among mammals warrants studies in human tissue. We describe a biopsy technique to collect living taste cells from the papillae on human tongue.
