Sludge Inoculum Preparation

Sludge inoculum preparation is the controlled collection and conditioning of microbe-rich sludge to provide a consistent starting population for biological experiments and treatment systems. The process preserves or adapts active microorganisms by mixing sludge thoroughly, adjusting its concentration or environmental conditions, and introducing it into a defined medium or reactor, where microbial metabolism drives target biological reactions. In biology and environmental biotechnology, prepared inocula support studies of biodegradation, nutrient removal, anaerobic digestion, and microbial community performance. Consistent preparation improves experimental comparability, helps researchers evaluate operating conditions, and enables reliable scaling of biological treatment processes.

Microbial sulfate reduction is a process of great importance in environmental biotechnology. The success of the sulfidogenic reactors depends among other factors on the microbial composition of the sludge. Here, we present a protocol to develop sulfidogenic sludge from hydrothermal vents sediments in a UASB reactor for reductive dechlorination purposes.
