Stringent Response Inhibition

Stringent response inhibition is the suppression of a bacterial stress-adaptation pathway that reallocates cellular resources when nutrients, especially amino acids, become limited. During starvation, alarmone nucleotides guanosine tetraphosphate and pentaphosphate, collectively called (p)ppGpp, accumulate through RelA and SpoT activity, altering transcription to reduce growth-related processes and promote survival; inhibition prevents or reverses these changes by targeting alarmone synthesis, degradation, or downstream signaling. In biology, studying stringent response inhibition clarifies how bacteria balance growth and stress tolerance and can support antibiotic development, infection research, and strategies that disrupt persistence or sensitize cells to treatment.

In Vitro Directed Evolution of a Restriction Endonuclease with More Stringent Specificity

Restriction endonucleases with new sequence specificity can be developed from enzymes recognizing a partially degenerate sequence. Here we provide a detailed protocol that we successfully used to alter the sequence specificity of NlaIV enzyme. Key ingredients of the protocol are the in vitro compartmentalization of the transcription/translation reaction and selection of variants with new sequence specificities.
