Ppgpp Effect

The ppGpp effect refers to the broad physiological changes triggered by the bacterial alarmone guanosine tetraphosphate, ppGpp, during nutrient limitation and other stresses. RelA and SpoT-like enzymes synthesize or degrade ppGpp, allowing its concentration to shift rapidly; ppGpp then acts with regulatory proteins and RNA polymerase to redirect transcription, reduce growth-associated biosynthesis, and support stress adaptation. This stringent-response mechanism helps bacteria balance survival with cellular growth and influences metabolism, ribosome production, persistence, and interactions with hosts. Studying the ppGpp effect clarifies bacterial regulation and can inform research on antimicrobial strategies and stress-resistant infections.

A Purification and In Vitro Activity Assay for a (p)ppGpp Synthetase from Clostridium difficile

Here, we describe a method for purifying histidine-tagged pyrophosphokinase enzymes and utilizing thin layer chromatography of radiolabelled substrates and products to assay for the enzymatic activity in vitro. The enzyme activity assay is broadly applicable to any kinase, nucleotide cyclase, or phosphor-transfer reaction whose mechanism includes nucleotide triphosphate hydrolysis.

The growth of radiolabeled bacterial cultures in microtiter dishes facilitates high throughput sampling that allows multiple technical and biological replicate assays of nucleotide pool abundance, including that of (p)ppGpp. The effects of growth transitions provoked by sources of physiological stress as well as recovery from stress can be monitored.

Flow Cytometric Analysis of Particle-bound Bet v 1 Allergen in PM10

Here, we present a protocol to quantify allergen-loaded particles by flow cytometry. Ambient particulate matter particles may act as carriers of adsorbed allergens. We show here that flow cytometry, a method widely used to characterize suspended solids >0.5 µm in diameter, can be used to measure these allergen-loaded particles.
