Substantia Nigra

The substantia nigra is a group of pigmented nuclei in the midbrain that helps regulate movement, motivation, and aspects of learning through its connections with the basal ganglia. Its pars compacta contains dopamine-producing neurons that modulate striatal circuits, while its pars reticulata provides inhibitory GABAergic output to thalamic and brainstem motor pathways. Loss of dopaminergic neurons in the pars compacta disrupts basal ganglia signaling, contributing to the tremor, rigidity, and slowed movement characteristic of Parkinson’s disease. Studying the substantia nigra supports research on motor control, neurodegeneration, brain circuitry, and treatments such as dopamine replacement and deep brain stimulation.

Here we describe a dissection and sample-handling approach to maximize results, and thus conclusions and insights, on dopamine regulation in the midbrain nuclei of the substantia nigra (SN) and ventral tegmental area (VTA) in rodents.
