Tetracycline Inducible Cells

Tetracycline inducible cells are genetically engineered cells that allow researchers to switch a target gene on or off using tetracycline antibiotics, such as doxycycline. In the Tet-On system, doxycycline binds a reverse tetracycline-controlled transactivator, enabling it to recognize tetracycline response elements and activate transcription; related Tet-Off systems use tetracycline to block gene expression. This conditional control helps separate gene activation from cell development and maintenance, reducing the effects of constitutive expression. Tetracycline inducible cells support studies of gene function, signaling pathways, differentiation, disease mechanisms, and drug responses in cultured biological models.

Here, we use retroviral transduction and concatemeric transfection to create a cell line that can express the components of a lentiviral vector (LV) in the absence of tetracycline. This LV encodes GFP and is pseudotyped with a glycoprotein, SVGmu, which is specific for a receptor on dendritic cells.
