Transit Amplifying Cells

Transit amplifying cells are rapidly dividing progenitor cells that arise from tissue stem cells and generate the specialized cells needed for growth, maintenance, and repair. After receiving signals that promote proliferation, they undergo a limited number of symmetric or asymmetric divisions before exiting the cell cycle and differentiating into mature cell types. This intermediate population expands the output of a small stem-cell pool while preserving long-term stem-cell self-renewal. Studying transit amplifying cells helps explain tissue regeneration in systems such as the intestinal epithelium, skin, and nervous system, and reveals how disrupted proliferation or differentiation can contribute to developmental disorders and cancer.

We present here a cell culture method for inducing mesenchymal-epithelial transitions (MET) in sarcoma cells based on combined ectopic expression of microRNA-200 family members and grainyhead-like 2 (GRHL2).
