Zone Of Inhibition Measurement

Zone of inhibition measurement is a microbiological method used to assess how effectively an antimicrobial agent prevents the growth of bacteria or other microorganisms. In a disk diffusion assay, an antimicrobial compound diffuses through an agar medium seeded with microbes, creating a concentration gradient; the clear area surrounding the sample, called the zone of inhibition, is measured to estimate antimicrobial activity. In biology, this method supports antimicrobial susceptibility testing, comparison of treatments, and evaluation of substances with potential therapeutic or preservative uses. Results depend on factors such as diffusion, microbial growth conditions, and agent concentration, so standardized procedures are essential for meaningful comparisons.

We describe the enzyme-linked lectin assay (ELLA) for measuring influenza neuraminidase (NA)-inhibition antibody titers in sera. The assay uses peanut agglutinin to quantify galactose residues that become accessible when NA removes sialic acid from fetuin-coated, 96-well plates.
