Crypt Pellet Centrifugation

Crypt pellet centrifugation is a laboratory method used to collect intestinal epithelial crypts as a concentrated pellet for downstream analysis. It works by suspending released crypts in a liquid and applying centrifugal force, causing the crypt structures to sediment while smaller particles and soluble material remain in the supernatant; repeated washing or centrifugation can improve sample cleanliness. In cancer research, the resulting crypt-enriched preparation supports examination of epithelial organization, stem and progenitor cell behavior, tissue injury, and tumor-associated changes. It can also provide starting material for molecular assays and three-dimensional culture models that help investigate intestinal tumor initiation and progression.
