Tumor Graft Isolation

Tumor graft isolation is the process of recovering tumor tissue or tumor-derived cells from an experimental graft while separating them from surrounding host tissue. In cancer research, investigators typically excise the graft under sterile conditions, remove non-tumor material, and preserve the sample through washing, mechanical or enzymatic dissociation, or direct tissue processing, depending on the intended analysis. The isolated material can support histology, molecular profiling, cell culture, transplantation, and assessment of tumor growth or treatment response. Reliable isolation helps maintain representative tumor features and generates reproducible samples for studying tumor biology, therapeutic efficacy, and interactions between cancer cells and their microenvironment.

In this report, we describe a protocol for isolating highly purified populations of leukocytes that infiltrate tumors. This protocol is adapted from the Miltenyi Biotech protocol to enhance yield and purity for isolating cells from complex tumor tissue.

Here, we describe techniques related to the isolation of neutrophils with anti-tumor properties and methods for monitoring anti-tumor neutrophil function in vitro and in vivo.
