Exosome Downstream Analysis

Exosome downstream analysis is the set of laboratory methods used to characterize exosomes after their isolation, determining their molecular cargo, surface markers, size distribution, and biological activity. Analysis typically combines particle characterization with vesicle lysis or RNA extraction, followed by assays such as immunoblotting, flow cytometry, PCR, sequencing, or functional cell-based tests to connect exosome composition with its effects. In cancer research, these measurements help evaluate how tumor- or stromal-cell exosomes influence intercellular communication, immune responses, invasion, and treatment resistance, while supporting biomarker discovery and assessment of exosome-based diagnostic or therapeutic strategies.

Exosome Isolation: A Technique to Separate Exosomes from the Plasma of Non-small Cell Lung Cancer Patients

2023

The plasma-isolated exosomes can be used as a non-invasive and repeatable way for the detection and follow-up of the biomarkers for early detection and disease progression. This video describes the protocol for the separation of exosomes from the plasma samples of non-small cell lung cancer patients, which could be used for DNA and protein extractions.

(2018)This video demonstrates an in vivo mouse model for inducing and analyzing acute lung injury (ALI) using intraperitoneal administration of bacterial lipopolysaccharide. The inflammatory cascade triggered by alveolar macrophages and neutrophils leads to epithelial and endothelial barrier disruption, allowing fluid and immune cells to accumulate in the alveoli. Alveolar...
