Lentivirus-mediated Gene Delivery

Lentivirus-mediated gene delivery is a method for introducing genetic material into target cells using engineered lentiviral vectors, making it valuable for stable gene manipulation in cancer research. The vectors enter cells, reverse-transcribe their RNA genome into DNA, and integrate the transgene into the host genome, enabling sustained expression in dividing and nondividing cells. Researchers use this approach to express tumor-suppressor genes, silence oncogenes, label cancer cells, and modify immune cells for studies such as CAR T-cell development. Its durable delivery supports functional studies, disease modeling, and evaluation of potential cancer therapies.

In this video, we demonstrate a method to perform transduction of shRNA lentiviral vectors to obtain stable knockdown cell lines in immortalized hematopoietic stem and progenitor cells (iHSPCs).

Acufection-Mediated Delivery of Immune-Related Genes into Mouse Skin Tissues

The video demonstrates a method for directly transferring the interleukin-15 encoding plasmid DNA into the skin tissue of an interleukin-15 deficient mouse model through acupuncture using microneedles. These microneedles generate micropunctures in the outermost skin layer, facilitating the absorption and transfection of plasmid DNA into the epidermal cells, including keratinocytes, within the skin tissue.

Source: Eggers, C. H. Phage-Mediated Genetic Manipulation of the Lyme Disease Spirochete Borrelia burgdorferi. J. Vis. Exp. (2022).This video demonstrates the transduction of genetically modified Borrelia spirochetes using polyethylene glycol–precipitated, non-infectious bacteriophages carrying a kanamycin-resistance plasmid. After incubation with the bacteria, the culture is plated on antibiotic-supplemented medium. Only cells that receive the plasmid survive and form colonies. These colonies...
